Tritium Labeling

Tritium is the appropriate isotope when choosing labeled ligands for studies involving receptor binding or other systems that require a protein with properties identical to those of an unlabeled protein.  Substituting tritium for hydrogen atoms creates a product that is virtually identical to the parent molecule, but with a radioactive tracer.  Increased specific activity is achieved by increasing the number of tritium atoms per molecule, or by labeling multiple amino acids within a peptide.

With its high specific activity, relatively short half-life, low energy emissions and relatively low cost, tritium is often considered the ideal label.  Detection is reasonably quantitative when using a liquid scintillation assay coupled with the fact that scintillation cocktails are non-toxic.   Additionally, the tritium decay product is non-radioactive and low amounts can often be disposed of through standard waste removal systems.

In the laboratory, tritium does not require shielding or special storage.  Tritiated compounds are stable and store well under normal lab conditions.  The ease of handling greatly increases the locations where labeled materials can be used including biosafety cabinets and on the open bench.

Tritiated molecules have the same biologic and physical properties as their non-labeled counterparts.  Assays can be undertaken using physiologic concentrations of materials, reproducing conditions most like those encountered in vivo.  Concentrations of labeled proteins are easily titrated using non-labeled proteins without adjustment for activity, potentially increasing sensitivity in certain assays.

With over 80 years of experience in radioisotope labeling, the IsoSciences fast turnaround tritium labeling facility stands out as one of the most experienced labs in the industry. In particular our experience has been substantially with multiple nuclei; 3H, 14C, 125I, 11C, 18F, 77Br, and 195Au HHH. Incorporation of these radioisotope nuclei has been broadly used in both academic molecules but also in molecules considered proprietary within the pharmaceutical industry. Importantly, our radioisotope team also has the full synthetic support of our world renowned stable isotope synthesis lab, adding many advanced degree chemists with specialties in sugars, heterocyclics, annulations, natural products, APIs and toxin syntheses.

IsoSciences recognizes the importance of correct equipment in order to undertake the broad range of tritium labeling and functional group conversions necessary to generate the final molecules required by our partners. Core equipment available at our Monmouth Junction (NJ) facility includes:

  • RC Tritec Tritium Manifold
  • Agilent 1100 (TQD) HPLC with a Packard Radiomatic 150TR Flow Scintillation Analyzer
  • Packard Tricard 2100 TR Liquid Scintillation Counter
  • Thermo TSQ Quantum Classic LC/MS/MS

Synthetic capabilities are a primary consideration in the IsoSciences’ tritium labeling facility. However we have placed an equal importance on the analysis tools available to our staff. As more labs look to outsource their tritium labeling work there has been a greater emphasis on generating complete support documentation that allows a partner to regularly accept material with additional QC analysis. As such we have developed a comprehensive set of analytical tools to provide tailored data packages. Our analytical tools include:

  • NMR – Varian 300MHz (multi-nuclei)
  • HPLC/GC (including prep HPLC)
  • Mass Spectroscopy
  • Agilent LC-MS
  • Waters Acuity LC-MS
  • GC-MS

The extensive experience in tritium labeling found at the IsoSciences radiolabeling facility allows us to undertake the many chemical conversions and protections necessary to complete a very broad variety of synthetic projects. We recognize that not all tritium labeling projects are simple catalytic tritium exchange chemistries and have developed equipment and resource capabilities that allow us to  complete everything from the complete synthesis of precursors, to simple tritium exchange reactions to more complex tritium incorporation as dictated by molecule limitations or client needs.

IsoSciences’ radioisotope synthesis team has experience with multiple nuclei; 3H, 14C, 125I, 11C, 18F, 77Br, and 195Au HHH and the incorporation of these radioisotope in pharmaceutical related chemistries and broader, general molecules. All of IsoSciences radio labeling staff have 15-20 years, are internationally recognized for their work and have substantial experience working within major pharmaceutical organizations.  The support received from the stable isotope research and development lab also adds support and significant synthetic depth and knowledge. The stable isotope team includes many advanced degree chemists with specialties in natural products, annulations, sugars, heterocyclics, APIs and toxin syntheses.

While it is not possible to list all the chemistries that can be undertaken in the tritium labeling facility, a representative list of important tritium incorporation chemistries includes:

  • Dehalogenation
    •  Using tritium gas
    • With sodium borotritide/palladium acetate (in presence of another Pd/C reducible group)
  • Double bond/triple bond reduction using tritium gas
  • Reduction using sodium borotritide
  • Methylation using CT3I
  • Tritium/hydrogen exchange
    • Tritium gas and Crabtree's catalyst/Rh black/other catalyst
    • With tritiated water and a catalyst (such as Platinum, Tris(triphenylphosphine) ruthenium chloride, etc)

When it comes to custom tritium labeling, the IsoSciences team recognizes the importance of quick turnaround to aid our customers’ project timelines. Keeping costs down is equally important in order to allow all our clients the ability to pursue the maximum number of critical R&D projects. However, above all IsoSciences’ team recognizes the extreme importance of generating and supplying high quality material with quality control and analytical data that fully supports not only structural determination but also the tritium incorporation placement and concentration. As many major pharmaceutical organizations are looking to outsource their custom tritium labeling needs, these organizations still face one recurring problem – how to accept materials without causing a burden on resourced limited quality control groups.

Historically the custom tritium labeling marketplace has struggled to supply materials that underwent quality and structural analysis that would ultimately allow end customers to regularly accept tritiated materials based on the certificates of analysis supplied by the synthetic group that generated the tritium labeled material. IsoSciences works with clients to generate a comprehensive packet of analytical information tailored to the a specific partner that, after initial qualification and validation, allows them to accept tritium labeled material as if it were an internally produced material. IsoSciences has developed quality packets where clients have required orthogonal chromatographic techniques, a full NMR spectral analysis, sometimes just a simple specific activity measurement – of course, on many occasions the end analytical data requires many aspects of analysis to be deemed suitable for acceptance based on IsoSciences certificate of analysis.
